Introduction.

In this experiment we will be using a bright field microscope to identify or observed bacteria from water taken from a pond, yeast, Crithidia fascculate smear, and, Fushsin stain, separate smear. See appendix 6-7.

A microscope is an instrument use to enlarge small specimen that cannot be seen or studied with the naked eye. The main purpose of a microscope is to magnify and increase the visibility of a small object. The magnification of the microscope is 10X engraved on the eyepiece. (1)

A bright field microscope contains two eyepieces, four set of objective lens and numerous knobs to help in focusing the images. With the help of the microscope, the shape and form of the bacteria will be identify. This experiment introduces the process of wet-mount slide that will be use thru out the course.

Lighting emitting from the illuminator is used to obtain the best resolution, and, is adjusted at each objective lens. The change occurs by rotating the iris diaphragm aperture to match the objective that is in use. (2) See Appendix 2

When looking at an object in the lens it appears sharper and sharper meaning that it has a great resolution: the distance between two points place as close as possible to be seen as one. (3)

The purpose of this exercise is how to safely use the different component of the microscope, and, how a microscope can help in measuring and identifying the various bacteria, eukaryotes, algae, and, protozoa that we will be studying from now on.

A stage micrometer, is made of slide with a precise divided scaled identification on the surface of it, it is also labeled, and, as been labeled a simple microscope due to the lack of objective lens. The micrometer is use to calibrate the divisions and measurement on the eyepiece reticle or eyepiece reticule. (4) See Appendix 1 and 3.
