Proper use of micropipettes

Micropipettes are equipment to transfer and measure very small amounts of liquid.

The tips of micropipette played a significant role in using different types of sample. Firstly, the micropipette is hold vertically and the plunger is pressed to the first stop and the tip is submerged into the container containing solution. When the tip is placed into the solution the plunger was released. Hence a partial vacuum is created in the tip. Thus, 1ml of solution which is equal to 1000µL is draw back up into the tip. In order to dispense the liquid, the plunger is pressed at first stop. The remaining residue in the tip is removed by pressing the second stop. Then the tip is ejected by the tip ejector to prevent any contamination to the solution.

The most commonly used tips are the large blue between 200 - 1000µL. The volume indicator digital is used to adjust the volume of solution to draw up into the tip of micropipette.

The accuracy and precision of micropipette can be enhanced by holding the micropipette vertically when immerse the tip into the solution to prevent the liquid from running into the pipette body. The tip should not touch the surface of container because this will allow certain liquids to flow out from the tip. Thus, there is a reduction in volume affecting the results. Next, the tip is not allowed to touch the bottom of the container which can lead to aspiration of air. Apart from that, the micropipette is used under consistent speed and pressure. The plunger is pressed smoothly at the first stop with a consistent speed which is then immerses the tip into the solution and the plunger is released at consistent speed.

Other than that, some precautions steps are taken when handling a micropipette such as do not drop the micropipette. The micropipette should not adjust below zero µL as well...
