METHODOLOGY

A. MATERIALS

The materials used in preparing the hay infusion are hay/ grasses free of insecticides and is gathered in a specific place, wide mouthed jar, and stockings.

During the experiment, microscopes, deression slide, piette and dropper were used.

B. PROCEDURE IN MAKING THE HAY INFUSION

In making the hay infusion, the following procedures were used.

1. A handful of dried grasses were folded and cut into smaller pieces to fit into the jar.

2. The cut grass was placed on the jar and water was poured until it the water was 2centimeters from the mouth of the jar.

3. The jar was covered with stockings to ensure ventilation and to avoid contamination from other organisms.

4. The jar was kept from direct sunlight for about _ days.

C. PROCEDURES IN THE CHARACTERIZATION OF PROTISTS

OBTAINING AND IDENTIFICATION OF THE SAMPLES FROM THE TOP OF THE

HAY INFUSION

1. A sample drop was obtained on the surface of the hay infusion using a dropper.

2. The drop was then placed on the depression slide and was viewed on the microscope at 40X and 100X magnifications.

3. The organisms found on the microscope were identified using the dichotomous key and diagrams provided by the instructor.

4. The means of motility of the organisms were noted by observing the presence of cilia, flagella, or pseudopodia.

5. The identified organisms were then sketched.

OBTAINING AND IDENTIFICATION OF THE SAMPLES FROM THE BOTTOM OF THE HAY INFUSION

1. Instead of using a dropper in obtaining a sample on the bottom part of the hay infusion, a pipette was used. The pipette was inserted at the deepest part on the hay infusion and obtains a drop of it.

2. The drop was then placed on the depression slide for microscopic viewing. The same magnification was used in viewing the specimen.

3. The identification of the organisms was done using the same dichotomous key and diagram provided b the instructor. The means...
