GRAM- STAINING

HYPOTHESIS: Gram staining is useful tool to differentiate bacteria types.

PURPOSE: The purpose of Gram staining is to differentiate the bacteria species whether it is Gram Positive which are purple in color, or Gram Negative which turns to pinkish in color. after staining and looked under the microscope you will be able to distinguish, what type of bacteria it is. This part of experimental process is important part of the research project to be able to identify the type of bacteria we are isolating for study.

MATERIALS: the materials used in this methods are: Sterilized inoculating loop, clean glass slide, the bacterial culture, crystal violet that act as the primary stain, gram's iodine, ethanol acts as decolorizer, and saffranin act as the counter stain.

SUMMARY: Gram staining is important in Microbiology it is highly useful in differentiating unknown bacteria whether it is gram positive or gram negative. On a clean glass slide, smear a portion of bacteria sample colony on the top part using a loop that have been sterilized under Bunsen burner. Use a wooden clip to hold the slide lay it slanted on a metal dish. Cover the smear with Crystal violet as a primary dye stain. rinse the smear with distilled water using the bottle wash. Apply the Gram's Iodine for 60 seconds, cover the slide with 95% ethyl alcohol for 15 seconds. rinse with distilled wash bottle water. Cover the smear with Safranin for 60 seconds as a secondary dye. briefly rinse with water, drain the excess water and air dry. look under the microscope to see result.

RESULTS: We be able to identify the type the type of bacteria based on its physical properties and color. The Upper right picture shows: cocci shaped, puple color Gram positive bacteria -Staphylococcus Aureus bacteria. Lower right picture shows: Rod shaped, purple color Gram positive bacteria- Bacillus bacteria. The last picture shows: a rod shaped, pink in color Gram Negative bacteria- E. Coli...
