Experiment: Understanding Aseptic Technique

Introduction:

The purpose of this experiment is to identify bacteria responsible for the infection that occurred in a child’s wound after being bitten by a puppy. Also, conducting tube-to-tube test.

Procedure:

A sample is taken from the wound of a dog bite victim. Phenol Red Glucose Broth is the medium used. Next, the phenol red tube is placed in the incubator at 37 degrees for 24 hours—results are noted. Next, we conduct the aseptic transfer tube-to-tube test. Phenol Red Lactose is the medium used. The Bunsen burner is turned on, the loop is sterilized, lids on the tubes are removed and inoculation is done. The culture is incubated for 24 hours at 37 degrees. The culture is removed from the incubator and results are noted.

Discussion:

The microorganism under study in this experiment is gram negative rods. The inoculation/contamination lights are red and green, once the caps were removed both lights turned yellow. Once the tube was removed from the incubator, the contents of the tube turned a yellowish color. I will take a stab at this and guess that the contents turned yellow because the bacteria reacted to the presence of glucose.
