THE LAB PLAN Before you begin the task of purifying your enzyme you need a plan. We grade it, but you use it – so you are writing it with an eye to it being helpful to you in the lab. A PDF of the Lab Plan is to be uploaded to Blackboard 9 before January 23. You cannot begin the lab without our approval of this assignment.

Your sources for the lab plan will be whatever you can find: chiefly the biochemical literature and the web. Whatever methods you find will have to be adjusted to our resources, and scaled to what we have available. If you find a protocol for purifying jack bean urease from

50 pounds of beans, you can safely assume that you will not have a sack of beans that large to work with. Use the following assumptions for scaling your protocol down to size:

• If you are using animal or vegetable source, assume you begin with 10 grams of material.

• If you are using a bacterial source, assume you begin with 2 grams of material.

• If you are purifying an enzyme from eggs, assume you will start with 2 eggs.

Your references will also describe the use of chromatography resins. It is likely that we will not have these precise resins, but have their functional equivalents. We will provide a list of chromatography resins available in the lab, as well as buffer components, etc. Your Lab Plan is to be written making reference to what we have available in the lab.
